Script Details

Multiple sclerosis (MS) is a chronic neuroinflammatory disease of the brain and spinal cord that is a common cause of serious physical disability.

Central nervous system (CNS) lesions are a hallmark of MS and are caused by immune-cell infiltration across the blood-brain barrier that promotes inflammation, demyelination, gliosis, and neuroaxonal degeneration, leading to disruption of neuronal signaling.

Initially, autoreactive CD4+ T helper cells are activated in the periphery.

Activated CD4+ T-cells increase the immune response by recruiting additional immune cells including: CD8+ cells, B-cells, granulocytes, monocytes, and mast cells.

These cells adhere to and then cross the blood-brain barrier endothelium.

In order to do so, activated cells produce pro-inflammatory cytokines (TNF-α and IFN-gamma), which activate adhesion molecules (LFA-1 and VLA-4) on the blood-brain endothelium.

Following entry into the CNS, CD4+ T-cells are reactivated by antigen-presenting cells.
