When a sample arrives at the office, it is immediately carried to the laboratory for initial evaluation and check-in. Initial readings include temperature and motility, abnormalities or deformities in the sperm cells, volume of the sample, and concentration of the sample. Due to the rigorous semen sorting process, sperm cells must first pass the initial check-in criteria to ensure that customers will receive a quality product.
Once the semen in evaluated, it is prepared for the sorting process. This includes adding a stain to the sample. The sample is incubated at thirty-four and one half degrees centigrade for sixty minutes. This incubation period allows the stain to bind to the cells, which aids in sorting.
X-bearing, or female, sperm cells have about four point four percent more genetic material than Y-bearing, or male, sperm cells. The more genetic material a cell has, the more stain it absorbs. Stained semen feeds through sophisticated sorting machines, where sperm cells flow single file past a laser. A computer measures the resulting fluorescence and identifies the cells as bearing either x or y chromosomes.A slight electric charge sends the sperm streaming into the appropriate container. Specially trained technicians closely monitorthe machines throughout the sorting process to ensure product quality.
Once sorting has been completed, the straws are filled and sealed. Male sperm are put in blue straws and female sperm areput in pink straws. Straws of fresh semen are packaged in clear straws, with color-coded plugs for males and females.
Straws are then frozen, or packaged for immediate transport if they are to be used fresh.
Once frozen, one straw of each gender is thawed in order to ensure the sorted semen is of acceptable quality and integrity. In addition, gender purity is tested. This ensures that the straws contain at least eighty seven percent of the desired gender sperm cells.
Once straws pass quality control, they are transported to the tank room, where they are inventoried and packed into canes in preparation for long-term storage at our facility or transportation to their final destinations.
The entire sorting process, from arrival at the laboratory to delivery at your doorstep can take as little as twenty-four hours for fresh semen, or forty-eight hours for frozen semen.
Conception rates of sexed semen rival those of conventional semen. Results of more than 400 inseminations reported by breeders from across the nation showed that sexed semen averaged seventy-six percent conception, ninety-five percent gender accuracy, and one point six fawns per doe.
