Preparation of DaunoXome 

This is a video for medical professionals on how to prepare a drug used to treat cancer patients. The style should be informative, but also conversational. The product is DaunoXome - pronounced Dawn Oh Zome.

The client has not decided on male or female talent. The read is pretty short - but does have some technical terminology. We are looking for you to record a paragraph or two as a demo of your voice and how you would approach the read. For the actual recording we will provide a finished cut of the video which will show you pace and give you all of the pronunciations.

This project will probably not be recorded until the end of May or early June. We are shooting the video next week.

Hello and welcome to this short video, showing you how to prepare DaunoXome® (Dawn O Zome) for intravenous administration.

DaunoXome® is indicated as a first line cytotoxic therapy for advanced HIV-associated Kaposi’s sarcoma. DaunoXome® is not recommended in patients with less than advanced HIV-related Kaposi’s sarcoma.

DaunoXome® should be administered intravenously over a 60 minute period at a dose of 40mg/m2,with doses repeated every two weeks.

Each vial of DaunoXome® contains daunorubicin citrate equivalent to 50mg of daunorubicin base, at a concentration of 2mg/ml. The recommended concentration after dilution is 1mg daunorubicin/ml of solution. DaunoXome® is a translucent dispersion of liposomes that scatters light to some degree. Do not use DaunoXome® if it appears opaque, or has precipitate or foreign matter present.
Before you begin, you will need to ensure that you have all materials available for the dilution. It is important to remember that DaunoXome® should only be diluted with 5% Dextrose Injection (D5W).

DaunoXome® must not be mixed with saline, bacteriostatic agents such as benzyl alcohol, or any other solutions.

Aseptic technique must be strictly observed in all handling since no preservative or bacteriostatic agent is present in DaunoXome® or in the materials recommended for dilution.

Wipe all materials to be used with sterile alcohol swabs.
DaunoXome® should be diluted 1:1 with 5% Dextrose Injection (D5W) before administration.

Withdraw the calculated volume of DaunoXome® from the vial into a sterile syringe, and transfer it into a sterile infusion bag containing the equivalent amount of D5W. The recommended concentration after dilution is 1mg daunorubicin/ml of solution. Note that the majority of adult patients will need more than one vial of DaunoXome® per dose.

As with all parenteral drug products, inspect the diluted DaunoXome® visually for particulate matter and discoloration prior to administration, whenever solution and container permit.
Remove the diluted DaunoXome® from the aseptic chamber and label immediately.

Diluted DaunoXome® should be administered immediately. If not used immediately, it should be refrigerated at two to eight degrees Celsius (thirty six to forty six degrees Fahrenheit) for a maximum of 6 hours.

• DaunoXome® should be administered intravenously over a period of 60 minutes.
• Do NOT use an in-line filter for intravenous infusion of DaunoXome®.
• Do not store partially used vials for future patient use. Vials are for single use only.
• As with all anticancer products, proper procedures for handling and disposal should be followed.
